Antibodies are needed for western blot, immunoprecipitation, and protein purification among other methods. It is difficult to purify a protein made recombinant (cloning the cDNA, expressing the protein, purifying the protein - see our Molecular Station article on Recombinant Protein Expression Systems). There are many custom peptide synthesis services which bypass all this! You can simply order a specific peptide of your protein, and then use this peptide to generate an antibody. There are many antibody vendors are service providers that will make antibodies using peptides that they will synthesize for you. This cuts time down and costs (time is money and who wants to express a cDNA!).

   Recommendations for Peptides to be made into Antibodies or Peptide Antibody Production:

  • Peptides for antibody production can be 15-25 aa
  • Longer peptides larger than 25 aa (amino acids) can be used, but it increases the cost
  • Very short peptides may result in a increase in antigens detected
  • If you want to use short peptides <10 aa : these should be selected with potential sequence homology in mind - such as protein family analysis
  • Shorter peptides (<10 aa) may present limited # of epitope
  • Use the Molecular Station Peptide Directory and the peptide bioinformatic tools linked there to determine if your peptide is antigenic.
